The Initiator Titration Model

Emeritus Flemming G. Hansen, Dept. of Bioengineering, DTU

The initiator titration model was formulated 30 years ago as an attempt to explain initiation control of the bacterial (Escherichia coli) chromosome replication. The model is based on the initiator protein DnaA and its binding sites, the DnaA boxes, in oriC, in the dnaA promoter, and at other positions on the chromosome. The model was analyzed by computer simulations using a stochastic approach. In silico experiments were designed to be compared with published in vivo experiments by varying input parameters. I will present the computer cells, the computer cultures, and some of the in vivo-in silico comparisons.
